TP53 Mutation Exist in Acute Lymphoblastic Leukaemia Patients

A recent study involving 830 adult patients has revealed that approximately one in ten patients with Acute Lymphoblastic Leukaemia (ALL) carries a mutation in the TP53 gene , the mutation is a powerful independent predictor of poor prognosis and a higher risk of relapse, regardless of other high risk genetic features..

Findings of the Study shows thats:

The study led by Dr. Caner Saygin from the University of Chicago Medicine, provides a Valuable point given Belows :

1. TP53 mutations were found in 17% of the adult ALL cohort.
2. Mutations are more common in older patients and those with Philadelphia chromosome-negative (Ph-) B-cell ALL, where incidence can reach 40-50%.
3. Median overall survival was significantly shorter for mutant vs. wild-type (WT) cases: 1.9 vs. 5 years for B-ALL and 1.6 vs. 9.5 years for T lineage ALL.
4. These patients show inferior responses to conventional chemotherapy.
5.  A Variant Allele Frequency (VAF) is more higher or eqaul to 45% is linked to particularly poor outcomes in older patients

Clinical & Treatment Implications for ALL

1. While novel therapies like blinatumomab or inotuzumab ozogamicin can improve initial remission, relapses are common and often involve “antigen escape” which is loss of CD19/CD20/CD22 markers..
